Summary of thesis

This research tracks the evolution of T cell receptor (TCR) repertoires during immunotherapy in non-small-cell lung cancer and melanoma patients. It compares TCR diversity and clonal expansion between responders and non-responders to immune checkpoint inhibitors (ICIs) using next-generation sequencing data, bioinformatics analysis, and functional laboratory assays. The research further identifies predictive biomarkers of treatment response and tumour-specific TCRs by performing a longitudinal analysis of tumour and blood samples. To detect clinically relevant TCR patterns and assess anti-tumour activity, the research employs machine learning tools and in vitro validation, respectively. The project seeks to improve personalised immunotherapy strategies.
